Although you can launch Web Inspector with option+cmd+i, there is no shortcut for Inspect Element. 

Using Firebug in Firefox, you can define a shortcut for the equivalent function of inspect element, which makes the process of inspecting an element's markup a simple case of hitting the shortcut then mousing over the element you wish to inspect. If Firebug is not already running, the shortcut for inspect element will open it, and then enter the inspect mode in one operation.

In Webkit, you first have to open Web Inspector with option+cmd+i, then click on the inspect element icon in the Elements panel. 

This may seem like a trivial thing, but as a front-end engineer I find myself inspecting elements on sites I browse many times a day, and the extra step in getting into inspect element mode is a real usability killer and one of the only things that is stopping me switching from Firebug to Web Inspector as my preferred working environment.
